The validation of novel target-specific radioligands requires animal experiments mostly using mic... more The validation of novel target-specific radioligands requires animal experiments mostly using mice with xenografts. A pre-selection based on a simpler in vivo model would allow to reduce the number of animal experiments, in accordance with the 3Rs principles (reduction, replacement, refinement). In this respect, the chick embryo or hen’s egg test–chorioallantoic membrane (HET-CAM) model is of special interest, as it is not considered an animal until day 17. Thus, we evaluated the feasibility of quantitative analysis of target-specific radiotracer accumulation in xenografts using the HET-CAM model and combined positron emission tomography (PET) and magnetic resonance imaging (MRI). For proof-of-principle we used established prostate-specific membrane antigen (PSMA)-positive and PSMA-negative prostate cancer xenografts and the clinically widely used PSMA-specific PET-tracer [68Ga]Ga-PSMA-11. However, subtle findings especially concerning small local recurrences can still be challenging to interpret and are prone to variability between different readers. Thus, we analyzed interobserver variability, detection rate, and lesion patterns systematically in a homogeneous patient population with low-level biochemical recurrence. Methods We analyzed 68Ga-PSMA-11-PET/CTs in 116 patients with status post-prostatectomy and PSA levels up to 0.6 ng/ml. None of them received ADT or radiotherapy beforehand. Während zunächst die Amyloid-Hypothese im Vordergrund der Diskussion zur Genese der DAT stand, wird dazu in den letzten Jahren zunehmend der Einfluss der pathogenen Tau-Ablagerungen diskutiert. Mehrere 18F-markierte β-Amyloid-Tracer wurden inzwischen arzneimittelrechtlich in Europa zugelassen. Sie sind somit verkehrsfähig und daher auch für niedergelassene nuklearmedizinische Praxen verfügbar. Radiopharmaka für Tau-Bildgebung mit der PET befinden sich ebenfalls in Entwicklung. In the current study, we synthesized DUPA-Pep having 2-[3-(1,3-dicarboxypropyl)ureido]pentanedioic acid (DUPA) linked via 8-aminooctanoic acid to two phenylalanine residues and chose 6-[ 18 F]fluoronicotinic acid 2,3,5,6-tetrafluorophenyl ester [ 18 F]FPy-TFP as a prosthetic group for coupling. [ 18 F]FPy-DUPA-Pep was obtained in a radiochemical yield of 48 7 0.9% (decay uncorrected) within 50 min with a chemical purity of 4 98%.

No. 1640 Objectives: For metabolic studies of tracers labeled with PET radionuclides and "classic... more No. 1640 Objectives: For metabolic studies of tracers labeled with PET radionuclides and "classic" radionuclides such as C-14 and tritium, on-line detection of β+/β-particles in radio-HPLC is essential.Since special Li-glasses (GS1) are available as internal solid scintillators, we evaluated the applicability of such a detector for metabolites labeled with N-13, C-14 and tritium, F-18, C-11 as well as in dual isotope studies. Methods: HPLC system was used with a Ramona as β-detector (raytest, Germany) in line. For calibration, 13NH3, [18F]FDG (140MBq/0.1mL), [11C]choline (108MBq/0.1mL), [3H]acetic acid (185MBq/mL), and [14C]aspartic acid (3.7MBq/mL),were used. Dilutions by a factor up to 10-100,000 were made. For analysis of 14C-labeled and 3H-labeled amino acids, SCX Partisil column was used. The system was also utilized for assessing the purity of commercially available [14C]proline, [14C]glutamic acid, [14C]aspartic acid, [14C]glutamine, [14C]alanine, [14C]urea (37MBq/mL), [3H]glutamic acid, and [3H]alanine. For dual isotope studies, a mixture of 13N-and 14C-amino acids was analysed by same HPLC conditions. Similar system was used for analysis of 13N-labeled amino acids in animal experiments. Results: Though certified with a purity of 99%, [14C]glutamic acid, [14C]aspartic acid and [14C]proline showed impurities of 20%, 10% and 72%, respectively. Calibration was carried out for 13N with 13NH3 (linear response interval:0.7-10kBq). A good recognition of an activity peak was possible at 40Bq for both 13N and 14C, 5kBq for 3H, 20Bq for both 11C and 18F. In dual isotope studies with 13N and 14C amino acids, the mixture of β+ and β-emitters was registered and after decay of 13N, analysis was repeated for measurement of 14C. That type of radiochemical standard approach allowed to detect even low activities (40Bq) of 14C.

For the reactions of the glutamine metabolism ammonia plays a crucial role and is therefore further investigated in the present study. Methods: The turnover of N-13-ammonia into glutamine and further products like urea was studied in SCID mouse xenografts of various cell lines (LNCaP C4-2, PC-3 and TRAMP). The animals were treated with intravenously administered N-13-ammonia for varying times and the tumors were dissected and homogenized afterwards. Insoluble cell fragments were separated and the supernatants were analyzed for N-13-metabolites via radio-HPLC. Blood samples were treated and analyzed similarly. Aim: Radioimmunotherapy with bone marrow seeking antibodies labelled with therapeutic radionuclid... more Aim: Radioimmunotherapy with bone marrow seeking antibodies labelled with therapeutic radionuclides reduces relapse-related mortality while causing only limited toxicity to non-target organs. Exact quantitative determination of activity tissue distribution and thus of organ exposure is still an unsolved problem because of limited image resolution and oversimplified quantitation models in planar gamma-camera imaging. In contrast, PET allows to determine tracer uptake quantitatively in target tissues. Therefore, a method to label antibodies with 18F via acylation was established. Materials and Methods: An automatic and remote controlled synthesis module was constructed to synthesize [18F]succinimidylfluorobenzoate as a prosthetic labelling group according to the method of Wester et al.
